Pan-RAF Inhibitors for Paediatric Low-grade Gliomas Offer New Opportunities in Targeted Therapy
نویسندگان
چکیده
Low-grade gliomas are the most common brain tumour to occur in childhood. The identification of mitogen-activated protein kinase pathway as near-uniform driver these tumours has led testing therapies targeted at this pathway, with promising early results. pan-RAF inhibitor tovorafenib is one recent agents be tested for paediatric low-grade gliomas, data raising prospect that we may dawn a new era management childhood tumour.

Targeted therapy in pediatric low-grade glioma.
Collectively, pediatric low-grade gliomas account for most brain tumors reported in children. Surgery is typically curable for operable lesions. However, more effective therapies are required for inaccessible tumors, both to overcome refractory disease and to minimize the toxicity associated with conventional adjuvant chemotherapy and radiotherapy regimens. Recent years have witnessed rapid imp...

CME ABSTRACT Objective:To evaluate the predictive impact of chromosome 1p/19q deletions on the response and outcome of progressive low-grade gliomas (LGG) treated with up-front temozolomide (TMZ) chemotherapy. Methods: Adult patients with measurable, progressive LGG (WHO grade II) treated with TMZ delivered at the conventional schedule (200 mg/m/day for 5 consecutive days, repeated every 28 day...
متن کاملDiffuse low-grade gliomas
Description : Diffuse low-grade gliomas (WHO grade II) are a sub-group of rare and heterogeneous primary brain tumors that usually occur in young patients living a normal life until the onset of a first seizure. A good understanding of the natural history of these gliomas namely: their steady progression, infiltration along white matter fibers and especially the risk of malignant transformation...
متن کاملPediatric low-grade gliomas and the need for new options for therapy: Why and how?
Pediatric low-grade gliomas are the most common tumors of the central nervous system in children, accounting for almost 50% of all childhood brain tumors. They are a heterogeneous group of tumors with different histologic subtypes. Most treatment studies address low-grade gliomas as a single entity, depriving us of histology-specific treatment outcomes. This is mostly due to a lack of understan...
متن کاملNew strategies in pediatric gliomas: molecular advances in pediatric low-grade gliomas as a model.
Pediatric low-grade gliomas (pLGG) account for more brain tumors in children than any other histologic subtype. While surgery, chemotherapy and radiation remain the mainstay of upfront treatment, recent advances in molecular interrogation of pLGG have shown a small number of recurring genetic mutations in these tumors that might be exploited therapeutically. Notable findings include abnormaliti...
